2007 Proof-of-principle for the construction and selection of DNA-routed libraries
In 2007, Harbury and co-workers described the DNA-routed chemical translation of an octamer peptoid library comprising about 107 compounds. Six rounds of “molecular evolution” on N-CrkSH3 (N-terminal SH3 domain of Crk protein) resulted in the identification of structurally related peptoid structures with micromolar affinities (best binder KD= 16 μM).
